Hi, I've recently done a FTIR analysis on empty and drug-loaded nanoparticles, and there are noticeable shifts in the peak intensities around 2950 and 1080. I was wondering if such a change could signal a polymer-drug interaction such as hydrogen bonding etc. The FTR spectra for empty NP (E0), drug-loaded (E6), and the drug are attached.

Hi, the change in intensity directly relates to the concentration of the respective functional group. Your peaks attribute to C-H. Interaction is manifested by shifting the peak to a different wavenumber.
Hi, thanks for the response. The peaks at the aforementioned wavenumbers are shifted slightly after encapsulation with the drug. Could this mean C-H is participating in making hydrogen bonds with the drug molecules (Donepezil)
What is the drug load and how did you record the spectra, ATR ? Generally IR is a 1 percent technique. There is no evidence of the drug in the loaded spectrum. In fact it looks as though something aliphatic CH based has been removed in the loaded rather than the EO spectrum. There is no evidence of H bonding in the spectra of EO or E6 spectra in the 3000 -2000 cm-1 region although there is in the spectrum of the drug.
An ATR approach was used to record the spectra. The drug used is Donepezil hydrochloride. Based on the in-direct technique for calculating encapsulation efficiency, some drug has been loaded into the nanoparticles. My question here is what caused this shift in intensity after loading the drug? If it cannot be explained by drug-polymer interaction, then what might be a possible justification?
I do not know what your nanoparticles are but the main bands look like a polyamide with some OH. Are they coated ? the main difference between EO and E6 is a loss of OH at 3200 and 1970 cm-1 plus loss of aliphatic CH3, CH2 in the 2900 and 1400cm-1 regions. There is no evidence of the bands from the drug even if it becomes less crystalline.
