This is the microscopic image of rat tail tendon with custom built Second Harmonic generation Laser Scanning Microscopy, (photon counting with Hamamatsu PMT modules).
But there were random-spread line-shape noises over image.
Which point should I examine first to get rid of that kind of noises?
As I recognized from your image, these black lines is not fully black, in these lines intensity of SHG signal is significantly lower then on the average on the image. In my opinion you should check stability of the output power of the laser source in time (average time of getting one image) must be constant. If laser source is OK you should check PMT module, place test SHG active sample (e.g. BBO crystal) and to detect SHG intencity in time (average time of getting one image) must be constant. For deeper understanding your problem I need more details (optical sceme etc.)
we had similar lines in a confocal LSM (Leica) - the problem for us was that the AOTF driver was failing... a replacement solved the problem...
