I got ELISA kit for a drug that I want to study from ABCAM and BioVision. Their protocols are exactly the same. Additionally, I'm pretty sure the reagents more or less come from the same manufacturer. Reasons being, those reagents and substrates are pretty standard for sandwich ELISA and come in a generic bottle.
However, ABCAM kit work significantly better BioVision when ran under exact same experimental conditions.
Does anyone how I can find out the differences between the two kit, be it the composition of the reagents or the way the plates were coated and blocked?
As far as I am aware, ABCAM gives you information on antibodies for ELISA kits, so you need to find what antibodies they provided. Differences in antibodies could be because they were raised against different portions of the same protein, for example (check the exact sequence against which they were raised), check reactivity (is it supposed to react to human or mouse?) etc. All the antibody information needs to be exactly the same, you need to check.
sensitivity of these two reagents may be different. one of these kit may just lose many points (results may be 0) if these points are below the threshold of sensitivity.
Try to switch reagent, run abcan on biovosion plate and vice versa. Or ask the company. Storage conditions (at the supplier)& batch difference may effect .
Try serial dillution for sample to determine the minimum amount which refer to the amount of coated plate
