I have purchased ubiquitin monoclonal antibody Ubi 1: Ubiquitin Antibody (13-1600) Thermofisher; I intended to check the quality of this monoclonal antibody and I run some brain nuclei at different developmental stages from E14 to P60. I also ran nuclei from HEK293 cells; this antibody was diluted at 1:500. Knowing that histone H2A (A24) gets ubiquitinated and that its molecular weight is at 24kDa, I expected to see a band at 24kDa that gradually increases through development (from E14 to P60). I did not observe any band at 24kDa. Instead, this ubiquitin antibody gives a signal at 37kDa and I have no idea what it corresponds to.
Has anyone come across this observation of finding a signal with ubiquitin antibody at 37kDa in nuclei of brain tissues or cells?
