Dear Sir. Concerning your issue about the concentration range that is use to build standard curve for ortho-nitro phenol. The linear determination range for o-nitrophenol is 1.0-25.0 µg mL-1 . For more details, I think the following below attached file may help you in your analysis:
First of all, you should see which concentration you have in your plant extract. In my case that I work in alkaloids plants, I prepare 1 mg/ml stock solution, and then dilute it to 1:2, 1:5, 1:10, 1:50 and 1:100. If I were you, I prepare the same concentration, and after seeing the components of your plants is in the same range. Then I use the same concentration. If it is not the same range, If it is higher, I prepare more concentratation, if it lower than 1:100, I prepare lower concentration.
