My cells express a receptor of interest and I would like to test if my cells also secrete the ligand for this receptor and activate themselves upon stimulation (autocrine regulation). Unfortunately, we were not able to detect the secreted ligand because it is not stable once secreted or immediately binds to the receptor. I would like to try a "trans-well assay by plating unstimulated and stimulated cells in two chambers separated by a membrane. If stimulated cells secrete the ligand, then this ligand would cross the membrane and bind to the receptors of the cells in the other chamber. I want to avoid cells crossing the membrane separating the chambers, I want only the ligand to penetrate. What commercially available chambers, plates, sets would you recommend? I would need to incubate the cells in the chamber at 37 degree TC incubator for at least 24 hrs. TIA!
