We are trying to desiccate BSA coated polystyrene microparticles (2-3 um diameter) in a plastic microchannel blocked with BSA/Casein + Tween 20. When we are trying to reconstitute the particles, the efficiency is fairly low i.e. very few particles are being extracted back into the reconstitution buffer (PBS & PBS + Tween 20).

The particle solution used for desiccation is a microparticles suspension in PBS with 0.1% BSA and 0.05% Tween 20.

On the contrary, when we pass the same particle solution through the channel, we are getting ideal result which is low binding to plastic surface. 

For desiccation, we fill the microchannel with the solution and leave it under vacuum with Silica Gel for 2-3 days until fully dried.

What do you think could be done to overcome the problem?
