I'm going to perform emulsion PCR using William's method but i'm confused of the specifications of each of the used reagents. for example; there are various types span 80 in sigma aldrich such as: 09569, S6760 and 8.40123 which one should i use? and the same for the other reagents.
Also, some papers use emulsion to aqueous ratio 3:1 and others use 2:1, which is better?
Polyoxethylene detergents can form peroxides in contact with air, these damage enzymes. I'd therefore buy peroxide-free qualities, buy only small amounts at a time and store them under N2, cool and protected from light.
For your application you would want chemicals that are nucleic acid free and do not inhibit the polymerase. Remember that there is no absolutely "pure" substance (Noddack 1934), but there are reagents pure enough for a particular purpose. The paper describing the method in question should point out any likely problems.
Engelbert Buxbaum thank you, for some of reagents there is no information about peroxides. they mention different viscosity and some published applications.
Benjamin THOHA Thomas There are few papers published for emulsion PCR especially with the above mentioned combination. in most protocols they mention the chemical name without the cat. no. and there are various types for each reagent. Also, I found 2 papers each one uses different type of span 80 and i don't know on what basis did the author choose the type?
