I think this depends so much on which journal you are reviewing for.

I used to review for the Mark Allen journals, which includes British Journal of Nursing. The readers were nurses or carers - for another of their publications - and the articles had both to be absolutely correct but also accessible to those who the journal targeted.

As I was a nurse practitioner working with both nurses and carers, I felt I understood what would be relevant to them.

Those whose work I reviewed were often also nurses or sometimes, carers.

However, I would not have felt nearly so comfortable.

There are not any particular characteristics or requirements. Some journals seem to allocate randomly (to people like me) who are well published on a certain area or topic. They foolishly think I am an expert or something. I'm not!

Some journals want you to apply and they will ask you to state your five best publications or something like that.

I think being selected or invited as a reviewer is a function of your expertise in the field or scope covered by the journal, aside the fact that you are a prolific publisher of articles in the related field with proven years of research and/or industrial experience. Sometimes, some journal publishers scout through online resources such as Research Gate, Google Scholar, Academia, Scopus, etc. for choosing eligible high-profile, reputable reviewers for their journals.

There are no specific qualifications needed to be a reviewer, but most reviewers will have a PhD. You certainly need to be able to prove you know the field of study well enough to be able to assess the novelty, quality, impact and importance of the research.
