I would like to prepare filamentous fungal biomass (Fusarium solani) to get a mycelium for biodegradation experiment. If anybody suggest me, would be useful.
Assume you're asking for an inoculation method. Use spores or you can also blend mycelium (e.g. http://www.nrcresearchpress.com/doi/abs/10.1139/x87-230?mobileUi=0.VgvtH_nF_sY)
If you have a cork borer you can use an agar plug from the plate you are growing them. It's a semiquantitative method but it's usually reproducible if you always use the same solid media, culturing conditions and age of the source solid culture. By the way, always take the plug from the border of the colony.
Thank you very much Xabier Vázquez Campos for your suggestion. If i use inoculum from solid medium, it would be a possibility to get agar when i check the dry weight after incubation.
I have worked with Fusarium flocciferum mycelium for biodegradation studies for many years; for biomass preparation, as mycelium or spores you can read my papers. If you have any doubt please feel free to ask.
