I made (cell-antibody) complex for target therapy purpose.
and I have trouble identifying whether the antibody is properly
conjugated on the cell surface.
I used ac4mannaz for cell surface glycoengineering, and antibody-dbco complex for conjugation (=azide + DBCO click reaction).
I have tried IHC and flow cytometry but the results were bad...
Can you recommend other methods for identifying surface antibodies?
Try to perform Biotinylation assay. it will help you to understand well about surface proteins. you need to be careful about what kind of protein you are targeting and you need to consider whether it has any internalization capability like EGFR. if you are targeting on EGFR, you must read about internalized proteins also.
You may also try a fluorescent antibody and fluorescence microscopy. Also, flow cytometry might be an option.
I think flow cytometry could be the better choice. Other option you have is an ELISPOT.
