I has a question regarding infection models- If you are inducing an infection model (bacterial) and then snap freezing the infected tissues in liquid nitrogen afterwards, does the pathogen affect downstream processing ? for example, with regards to western blotting, does it affect protein measurements with BCA assays? If so, how would you get around this issue? are there any protocols or steps you would recommend?
Snap freezing might not kill the pathogen. This means the samples should still be treated as potentially harmful until you have completed the protein extraction. Subsequent sample handling should be the same. The bacteria would contribute some protein towards BCA assays but the amount would depend on how many bacteria there are in comparison to the volume of tissue.
