A few suggestions state that always reviving THP-1 from cryo using it for few passage and doing experiments is advisable. But what differences may I expect when I cryo a batch of passage p-7 and revive it for experiments and in another case where I revive P-1 for my experiments. And why it is not that easy like other cell lines to continuously maintain THP-1.

For most of the monocyte/macrophage cell line, continuous passaging is not recommended. You can test your cell lines by stimulating them with LPS and their ability to generate nitrite in the media by simple griess assay. I had the same problem, a few weeks ago with my Raw 264.7 macrophage cell lines. But not between P1 vs P7. Usually upto P10 you should not expect any change. But prolong culture with >P15 has huge influence.
