I'm differentiating N2A cells using serum deprivation (0-2% FBS) and 20 µM RA. After 2 days differentiation is observable, after 4 N2A are still viable and have long neurites. However, on day 7 cells, especially the ones exposed to the lowest serum conditions die. I'm thinking about adding back regular media (EMEM + 10% FBS), however I'm afraid, that it will reverse the differentiation. What should I do to keep the cells viable after differentiation for further experiments. Ideally I'd want at least a week after differentiation.
