Exposure to eternal environment will definitely alter and contaminate the sample. During culturing of the sample there is probability of finding contaminants obtained from environment. This will effect specificity of the test. However, sensitivity of culture based tests may not be affected much by external environment in short term exposure but long term exposure is sure going to cause change in the microbes. It's highly difficult to determine it quantitatively as different environmental parameters have different detrimental effect on same sample itself and that also at varying levels.

That depends on the kind of enviroment. If the package is well-done and it fills every biosecurity requirement the probability is very low.

Is there any technological solution like "transportation container" available or developed to reduce such problem ?
