I have nano particle with polymer coated surface and I want to use those particles in vivo experiment. which test should be carried out before in-vivo experiment ?

Look at different in vitro tools for bio analysis of the polymeric nano-particles, this will give you some idea about the bio compatibility testing and there techniques, its correlation with different in vivo technique and further other interrelated aspects.
