I'd like to know what are some common ways for measure proliferative capacity of cancerous cells in cell culture. Say for example I manipulated cancer cells by telomerase inhibition, I'd like to see how their proliferative capacity was impacted after treatment.

You can use XXT assay to measure proliferative capacity of your cells.
- Un-manipulated- Control
- Manipulated- Test

As suggested above, the XTT and MTT assays only say about the metabolic activity of the cell. If you want to do the cytotoxicity impact of the treatments both of these methods work good but if proliferative capacity is your specification then I would suggest you go for either colony forming cell assay or FACS based cell cycle analysis.
