I'm interested in locating the myelin transition zone in serial cross sections of the mouse optic nerve. What antibody would you recommend for use in lightly fixed cryostat sections?
We usually use SMI-99, a mouse monoclonal antibody to myelin basic protein (MBP). It gives a robust signal, so usually a 1:1000 or 1:3000 dilution works well. It is available from Biolegend and several other suppliers.
thanks Peter. I spoke with my PI yesterday and we are also thinking we should look at a general RGC axon label -Do you think a neurofilament Ab would be best, or a pan-NF if it exists?
