I'm writing a college level laboratory activities manual for a general mycology course and I'm deciding on what activities to include in order to teach the essentials on mycology. I'd like to hear what people in other countries may advice. :)
I think you could fill lab manuals upon lab manuals with microscopy techniques. As far as various branches of mycology goes the microscopy techniques are the ones in common in all mycology field. After that depends on what kind of mycology you want to do.
I think you can include culture identification and some basic steps to avoid contamination. As mentioned in the earlier comment, based on organism character you can mention their sampling method also.
You have to go only with basics when you are writing such manual. So for this, firslty you have to write how to decontaminate the particular area (work bench), then up to the test what kind of test you are working for, and in the end you have to write microscopy (only in case of identification).
And whenever we have to write / update such kind of manuals then we should have to go with basics then look forward as this is for youngsters who are un-aware about the such tests...
Start with students possessing capabilities of aseptic technique and microscopy as well as basic knowledge of biology (taxonomy, cell structure) . The lab should be accompanied by lecture that acquaints students with the unique biology of fungi as distinct from other Kingdoms as well as the basic vocabulary of mycology. Lab should start with students observing in practice the basic structures of fungi and observation of examples of various taxonomic groups - ascomycetes, basidiomycetes, etc.so as to understand the differences. Address reproduction - sexual and asexual. Isolation from environments of complex microbial population. Then identification of species using microscopic and cultural techniques as well as moleculrt techniques if you have those capablities. Finally a section on fungi of medical significance.
Teach them basic cultivation techniques. The pf tek This will help them to understand the life cycle of fungi in a much more accessible and hands on way.
