I want to culture human HepG2 cells in serum free medium. Should I add all components of complete medium except FBS only?, and how long the cells will be alive in such free medium?

2) For the incubation time, it depends on the cell type however, most studies have reported that cells remain viable up to 16h-24h. Since HepG2 is an immortalized cell line, there would most probably be no viability problem. You could perform cell viability analysis in the basal medium at different time points, say 8h, 16h and 24h. A simple MTT assay would do the trick. Good luck.

Generally if you are avoiding FBS and you want to keep your culture serum-free, HPL works really well.
