I have very little sample for western blot. I am comparing protein expression with some genes which showed good results in qRT PCR. But due to lack of protein sample I am able to do only 3-4 antibodies by cutting the blots according to their molecular weight. If I do stripping method on these blot, are they valid to show in publication?
If your antibody is from a different species then you may be okay. Also, you can mention that in your manuscript as a note to readers. Make sure you develop the blot after stripping to confirm that you don't see any bands.
When I did membrane stripping, I have noticed, that the signal from the same protein reprobed with the same antibodies declines (while there was no signal after stripping in "no antibody" control). To my opinion this may be caused by partial stripping of the antigen along with antibodies from the membrane. I think, you'd better be aware of this.
