The easiest way would be to extract progressively PCL and HFIP with an organic solvent in which collagen is not soluble and then to analyze the insoluble residue. Good luck.

Dear Mahyar Sameti,

I agreed with Michel Spina, but you may try to extract the collagen by low concentration of acetic at chiller temperature as collagen will slowly degrade with temperature increment. Otherwise, you could extract collagen using certain kit. Good luck!
