In general, you need to know lamda max ( λmax) (the wavelength in which a substance gives maximum absorbance) for the analyzed substance to be able to detect the difference in absorbance (the decrease in absorbance is relative to decrease in concentration of the substance to be eliminated).

- Perform an absorbance scan for the tested solution using the wash solution as blanc (absorbance = 0.00).

- Absorbance scan means absorbance under different wavelengths (280 nm to 780 nm is sufficient).

- If the tested solution is concentrated (the absorbance is above the absorbance limit), it must be diluted.

- λmax is the wavelengh in which you get maximum value for absorbance.

- When λmax is determined (eg 480nm) set the wavelength of the spectrophotometer at this wavelength.

- Then, you can measure absorbance of the wash result. The value approx. 0.00 reflects the disappear of substance to be filtered.
