If iam doing extraction, say using seperatory funnel , and in each wash i need to check for a certain compound that it is going to zero concentration . using UV -VIS and it is no more there in the wash , how can i go abou that ,, with steps please
Use a 2 vial uv analysis--first vial is solvent blank, the second is a volumetric sample of unknown. Each consecutive wash would have the same volume removed for each run. However, first you need to determine what a "zero" result means. Namely, is the limit of detection low enough for the "zero" to mean that what is left is low enough for your needs. Your instruemtent's noise level will determine the "zero" limit. If it is necessary to remove 99.99+ percent plus, you might want to use a different method.
You may use TLC screening of original solution against solution or sample after wash using suitable mobile phase and check it on Uv. You can check if your certain compounds were washed or still present
