I am interested in quantifying a range of steroids by LC/MS (C18 column with 70% methanol and 30% 25 mM ammonium acetate mobile phase, using atmospheric pressure electrospray ionisation) and usually have the strongest signal from the sodium ion adducts. To make the signal from sodium adducts more consistent, I've been including 1 mM Na+ in the mobile phase, but this causes a gradual decrease in sensitivity with each run. Sensitivity is restored after a period of idle time. At this point I only have external standards and so need the sensitivity to remain constant throughout a batch of samples and standards. Any suggestions about what is an appropriate concentration of Na+ for this approach or if there is another approach altogether for obtaining consistent steroid signals?
