Our incubator for primary cultures is broken but I need to isolate some islets for both qPCR and estimating the insulin content. Has anyone performed either of these techniques on freshly isolated islets? I'd greatly appreciate any feedback

Both of those things are OK to do on freshly isolated islets. The transcriptome does change after an overnight culture though, so if this is something you're planning on doing multiple times then just be sure to be consistent in your methods!

Yes but, it depends on your research question. Usually you keep them in culture for one or two days to recover from the isolation procedure, and make sure any experimental treatment afterwards gives you a consistent effect. You might run the risk of not getting a proper result due to the stress induced by the isolation procedure, make sure you are looking at the right things.
