I've encountered problem with large but scarce PDL crystals and spotted coating after 1hr of 0.02mg/ml PDL in PBS incubation, PBS washed and dried. Previous coating from same PDL stock resulted in the normal thin and even whitish layer. I've repeated with 0.1mg/ml or with new PDL stock but same problem occurred. Anyone have relevant experience? This is a charge enhance and ca be used for many metal coating surface. High PH will be required to add binding the ELISA plates. As I understand from my friend, I think timing one hour you are addressing may not be sufficient. There are reference you can take for your further study.
In case of uneven coating, the glass slides can be pretreated with 1 mM magnesium acetate or any suitable acetate for 3 hours and then rinsed well before coating or they may be acid washed (hydrochloric acid or sulfuric acid). No need to sterilize slides coated with poly-D-lysine by autoclaving; simply exposing the slides to UV light overnight is enough.

Use poly-d-lysine in water not in PBS. Wash in water then dry.
