Or a positive selection of my cells? Note: I normally perform erythrocyte lysis but when I try fycol separation I also get a lot of debris. Also positive selection with CD45 ab's isn't very effective. Any hints?
How many times do you wash your cells after erythrocyte lysis. I have found that the debris is due to platelets. So 2-3 washes with medium or buffer followed by low speed centrifugation (~900-1000 RPM) will normally remove them.
