Hello, I would like to ask about the preparation details of the dissolution scheme for animal experiments of Trametinib (HY-10999, MCE):
1. Is 0.5% HPMC (hydroxypropyl methylcellulose) calculated by w/v? Does it need to be heated or stirred overnight during dissolution?
2. Is there any particular order for adding 1% Tween-80? Does it need to be dissolved in saline first and then mixed with HPMC?

The solvent preparation operation is as follows:
1. Preparation of 0.5% HPMC (w/v) solution: Weigh 5 mg of hydroxypropyl methylcellulose (HPMC, model HY-A0104), add 1 mL of ultrapure water, and use vortex oscillation or magnetic stirring to completely dissolve until the solution is uniform, colorless and transparent (no particles remain).
2. Add 1% Tween-80: Add 10 μL of Tween-80 (1% by volume) to the above HPMC solution, continue vortex oscillation to mix, and finally form a uniform and transparent mixed solvent system.
