I have beginning work in a new lab where I never use the cryostat (but I did it a ton of times in my previous lab). I cut mouse visual cortex at -20 and -18 with sections of 20 um. The problem is that when I rise the antiroll my sections roll to much and it is impossible to stick them to the superfrost slide. Do you have any suggestions?

I've had a similar issue, what worked for me was skipping anti-roll plate and using a brush to stretch the sections. Or you can try with anti-roll plate, but just not cutting all the way through, leave a bit of OC at the end, then use a brush to gently roll it out and mount directly ( the warm slide will melt the rest of the OCT). Check the angles of your cryo stage as well, try to adjust the blade/stage and chuck holder handle to something that works for you.

when my sections roll too much I leave them under the anti-roll plate and I get ready with 2 fine tip brushes. As soon as I lift the anti-roll plate I use the brushes to flip the section and flatten it on the other face (similar to what you do with a sheet of paper that has been rolled on one side for too long).

Let the section sit under the roll plate for a few seconds, then the section should "relax" enough that it won't roll up as much. You can also use a fine brush to gently unroll the section then try to pick up the section as quick as possible. Sometimes the roll plate can be slightly adjusted so the section won't roll up as much.
