Hi all,

i  am trying to do transformation with plasmidic extract from antibiotic resistant Klebsiella to DH5a cells. The plasmids should carry the resistance genes, so I am trying to see if this resistance could be transferred by transformation in E.coli..I am struggling because I have been repeating the transformation more times but I had no colonies at all. I don't know if this is due to the fact that the plasmids are quite large and probably at low copy number. Anyway, I was suggested to have a go with transformation of all the DNA extract (that includes plasmid and chromosome DNA). This is because multiple mechanisms  (not only the resistance genes on plasmids) and other genes could be responsible for causing the resistance..so, transforming with all the DNA could give the chance  to have transformants that are antibiotic resistant. Does this make sense for you? Does this sound unusual ?

Any suggestion (also about different techniques) would be really appreciated.
