I transformed XL2 blue chemically competent E. coli with 1 µl (10 pg/µl) or 10 µl (1 pg/µl) of pUC19 and got completely different results: about 100 colonies for the transformation with 1 µl and no colonies for the transformation with 10 µl. The DNA is identical, I took 1 µl from the same tube and either added it on the cells or diluted it with DEPC-treated water to 10 µl. Nuclease activity shouldn't be a problem, since I used DEPC-treated water and incubated the diluted sample only a few minutes on the cells on ice before the heat shock.

Since the number of plasmids is identical I would expect a similar number of colonies (within the variation of such an experiment). Does the dilution decrease the transformation efficacy so much? Does anybody have similar experiences?
