I have been trying to react 20bp ssDNA strands containing a 5' amine group with a commercial SMCC linker. I am using a FAM-NHS molecule as a control and this has been able to react just fine. The yield is increasingly better with higher PBS concentrations. With 10x PBS giving me a yield of about 50%. These reactions were all at pH 8.4, however the FAM-NHS works but the SMCC linker does not. I have tried different volumes of reaction, different temperatures, different levels of shaking and some pHs (between 7 and 9) and a couple of buffers. If anyone has worked on anything similar please let me know if you have any tips on what I could try next with the linker. Thank you!
