I am performing transformation for the L4440 (2790 bp) plasmid into DH5 Alpha, but no colonies are forming. I performed the same procedure using PUC19 and observed colony formation. My competent cells are working, and based on the colonies formed, the yield is high. However, when I perform the same procedures for L4440, I am not getting any results. How can I resolve this issue? I am using the following protocol for transformation.

1. Thaw competent cells on ice.

2. Add 5 μl of ligation product to the thawed cells.

3. Incubate on ice for 30 min (do not vortex or shake during this step).

4. Incubate at 42°C for 1 min, then on ice for 2 min.

5. Grow in SOC medium at 37°C at 200 rpm for 1 hour.

6. Spread the growing cells onto an LB plate containing 100 mg ampicillin.
