Does anyone knows the right solvent for dipeptide recrystallization for coupling between hyppurique acid and proline methyl ester hydrochloride, I have tried methanol and the ethylique ether but the results aren't with good yields.
Dear Hajar Benrzeil many thanks for sharing this very interesting technical question with the RG community. Your compound is rather polar, thus is will certainly be soluble mainly in polar organic solvents. Of course there are very many solvents and solvent combinations possible. You can for example try a diffusion method where you dissolve your compound e.g. in DMF and let diethyl ether diffuse in. A potentially useful single solvent which often provides good results is acetonitrile. You can try to dissolve the compound in hot acetonitrile and let the solution slowly cool to room temperature to (hopefully) result in crystal formation. In any case it would certainly be helpful to make a few quick solubility experiments before you start. Just place a few mg of your compound in small vials and add solvents of different polarity to find out what the solubility in these solvent is.
For a good overview about the various crystallization techniques please have a look at the attached very useful guide.
Good luck with your crystallization experiments and best wishes, Frank Edelmann
I'm not sure if the answers given here are really useful for you. As mentioned in my first answer, you should perhaps make your own solubility tests and then choose a suitable solvent or solvent combination.
