Mobile phase: Acetonitrile:phosphate Buffer(20:80), pH 7, 0.1% triethylamine added
Column : 4.6*150 mm c18 (end capped), Guard column (not more than 50 injections upto now) attached
Temperature = 30 C (fixed for every experiments)
Flow rate=1 ml
Detector : flourescence
So, during every new experiment, the retention time of the last basic compound changes (normally increases). So, can anyone suggest how to solve this problem in HPLC?
[ I wash column after finishing experiment by distilled water (filtered) for 15 min, 50% methanol for 45 min and 100% methanol for 30 minutes]
