Dear friends. When I tested progesterone receptor coregulators during an oestrus cycle in a cow, I got some surprising results. The peak of protein expression occurs before the peak of mRNA expression. It would be more logical if I observe the opposite effect. Is it possible to explain this effect?
This is a much observed and discussed phenomenon. Due to things like RNA half-life, RNA-turnover, etc, it is not always possible to correlate message (RNA) level expression with actual gene expression (protein). One would expect them to go hand in hand, but you can frequently down-regulate an RNA without seeing and effect on protein expression, and same with over-expression.
This effect requires users to validate their RNA-seq or other message-level analysis data. RNA expression modulation may not be sufficient by itself to prove protein expression modulation.
