Despite this, lipofectamine it is not the best vehicle to transport your miRNA inside the cell. Did you try already the XFect from TakaraBio? The option from Takara has a lower cytotoxicity, perhaps could be an alternative to you.
The lipofectamine is constituted of 3:1 mixture of DOSPA (2,3‐dioleoyloxy‐N‐ [2(sperminecarboxamido)ethyl]‐N,N‐dimethyl‐1‐propaniminium trifluoroacetate) and DOPE while XFect is formed by biodegradable nanoparticles.
In my doctorate I used the RNAimax from Thermo Fisher Scientific to do my transfection, but instead of in KG1 cells, was in NRK52E cells.
K562 is among the myeloid leukemia cell lines that we transfect most frequently. We use electroporation and the cells recover after 2 days with peak expression of target around Day 3 post-transfection.
