I am not sure whether or not this question was asked. I wonder can I induce expression of my protein with tetracycline or doxycyxline while seeding cells instead of one day after seeding cells?
It depends on several factors, in particular what protein, what cell type and the treatments.
In general I'd advise against it, because the seeding already causes cellular stress and the antibiotics increases it.
Even if the cell doesn't die because of it, the expression could be altered and the controls might not be reliable because the cells haven't reached homeostasis after seeding.
I would like to add my own answer in case someone may look for the answer of same question in the future. I tried to seed cells with tetracycline to induce expression of target protein (the lac operator system). Next day, I measured expression of my target protein through luminescence (my protein is tagged with HiBiT). I got signal up to 7 order of magnitude. (Signal level depends on your target protein, some proteins showed 5-6 order of magnitude). In this way, you can save 1 day for simple biochemical assays.
