I'm currently preparing a liposome loaded with Ascorbic acid and doxorubicin both in aqueous media. I have prepared liposomal ascorbic acid using microfluidics and was planning to add the doxorubicin but I wanted to to use another hydrophilic mock drug before purchasing the drug because it's expensive and I have a limited project budget.
I need to measure the entrapment efficiency and the effect of the ascorbic acid on the liposome loading capacity using a colorimetric assay. I thought of methylene blue but the ascorbic acid reduced the dye changing the colour (colourless) and led to the coalescence when both dissolved in PBS. I was hoping if you have any suggestions or if you can direct me.

I can see why you are trying to combine doxorubicin and ascorbic acid within a nanoparticle delivery system. I assume the logic is to mop up any oxygen free radicals generated by the cytotoxic drug (Am I correct?). However, you should remember that mixing cytotoxic drugs with other compounds can be tricky due to changes in the pharmacokinetics and pharmacodynamics. I would, therefore, suggest you conduct experiments to study any potential interactions between these two drugs before you start working on developing the nanopharmaceutical.

The best hydrophilic model drug is calcein which is fluorescent and cheap!
PS: Curcumin is hydrophobic and is not suitable for loading into the liposome
