Hi, I am having a Moraxella culture in blood agar and I wish to preserve it in glycerol stock. I tried to grow the culture in nutrient broth and also in trypitc soya broth but there was no growth after 24 hours. So can any one suggest a broth that can be used for its cultivation in bulk
Moraxella spp. grows well on sheep blood agar incubated at 37 degrees C for 24-28 hours. Don't have any information on broth.
Found a paper on how BHI (Brain Heart Infusion) broth works well with Moraxella. After which, it can be cultivated on Chocolate Blood Agar.

A broth that is supporting Moraxella spp. growth is Tryptone-yeast extract broth, however if for the purpose of stock culture you can harvest from the Blood Agar, suspend in tryptone soy broth and transfer in a glycerol tub.
Moraxella is a pilus (fimbria) producing bacteria. You will notice, typically in 48 hours on a solid face, the colonies corrode the agar and this is associated with fimbria production.
Mutants unable to produce this adhesin - prior to the initiation of disease - are avirulent.
In culture Moraxella undergoes colonial dissociation producing colonies which lack pili. This is an other reason why you want to select corroding colonies.
