I am looking for substrates (oligo- or polysaccharides) to test the activity of a putative ß-D-galactosidase. Do you have any recommendations? So far I only have pNP substrates.
I should have phrased my question better. Sorry for that!
First of all, I am working on a exo-galactosidase which is active on ß-D- but not alpha-D-galactose-pNP. Therefore, I am calling my enzyme a ß-D-galactosidase.
I am happy with the pNP substrates (I do not need the fluorescent substrate) and I could test X-Gal and lactose BUT I would like to have different kinds of substrates in order to determine substrate specificity better (similar to what people do with exo-fucosidases using blood and milk oligosaccharides varying e.g. in linkage and DP).
I was looking in papers and the website for different chemical suppliers for potential substrates (other then lactose, XGal and pNP) but I could not find any. That is why I was posting the question here :)
I also do not know which substrates my enzymes might degrade since it comes from a marine bacterium. That is why I have not tested lactose yet. I am not looking for chromogenic substrates in particular but anything with terminal galactose and I already checked Carbosynth and Elicityl.
