I am planning to start antioxidant enzymatic assay of spinach plant. i will do this assay from the stem part of the plant. Can anyone suggest me the standard protocol for this assay
Antioxidant activity was determined by 1, 1- diphenyl-2- picrylhydrazyl (DPPH) free radical scavenging method. A 0.5 ml of plant extract was added to 2.5 ml of a 0.004 % ethanol solution of DPPH. Absorbance at 517 nm was determined after 30 min and the percentage inhibition activity was calculated from [(Ao-A1)/ Ao] ×100, where Ao is the absorbance of the control (DPPH solution) and A1 is the absorbance of the extract/standard.
