I tried to dissolve ethyl acetate extract in water but it does not dissolve. If i dissolve it in another solvent, the absorbance varies drastically. Do i need a separate calibration curve of gallic acid for that particular solvent?
Extract dissolved in DMSO have been used in phytochemical analyses and phenols are a chemical class under phytochemical analysis. So, dissolve the extract in DMSO, then determine it phenolic content.
What is the assay you are using to determine the total phenol content?
There are many solvents used for the extraction of total polyphenols, especially when the Folin-Ciocalteau method is used for the determination of total polyphenols. such as ethanol and methanol.
Folin-Ciocalteau assay does not measure the total phenolics contain. It measures the total antioxidant capacity (TAC) and works in water. It can't be used in other solvents.
