That is a great question. The only definitive ways to measure what you have asked are spectroscopic. The reason is that extraction methods, especially sequential extractions, can modify the natural partitioning between soil phases. There are authoritative publications to this effect.
Aqua regia digestion method as an effective and useful method to determine the concentration of heavy metals. The Aqua regia method may be used to measure Cu, Ni and Zn in soils affected by human activities and therefore may be suitable to analyze regularly the heavy metal contamination in soils. Aqua Regia solution is effective in dissolving sulfides, phosphates and many metals and alloys such as Pt, Au and Pd. Determining the releasing part with Aqua Regia provides valuable information on the long-term presence of heavy metals in the soil.
