To prepare a dopamine-modified sodium hyaluronate hydrogel, first create a dopamine-modified hyaluronic acid (DA-HA) by bonding dopamine to sodium hyaluronate. Crosslink this derivative using sodium periodate, or alternatively, crosslink double-bond modified hyaluronic acid and graft dopamine onto it. Combine the crosslinked and dopamine-grafted hyaluronic acid with water or solvents to form the hydrogel.
1) 0.5 g HA + 50 ml PBS (pH: 5) + 0.25 g NaIO4 : 5 h mechanical stirrer in darkness / RT (0.5 ml ethylene glycol / 1h) , dialyzed against ultrapure water for 2 days and lyophilized.
2) 0.3 g AHA + 30 ml PBS (pH: 5) + 0.3 g DP : 10 h mechanical stirrer in darkness / RT , dialyzed against ultrapure water for 2 days and lyophilized.
3) DAHA macromers (5%, w/v) ( 0.01 g DAHA in 0.2 ml DI ) + 0.2 ml NaOH (pH: 8.5) ( contains 0.0111 g NaIO4)
No changes were observed and no gel was formed.
