I am thinking that sodium acetate would be equivalent to potassium acetate in a neutralization buffer to be used after alkaline lysis of bacterial cells for plasmid extraction. The recipe calls for a bit of 5M potassium acetate solution, but all I have on hand is sodium acetate. I am wondering if a 5M sodium acetate solution would be equivalent?

it looks like a 3M sodium acetate solution might be more appropriate.

The plasmid extraction by alkaline lysis involves addition of sodium dodecyl sulfate to lyse the cells and sodium hydroxide to denture the DNA. Post that, you might want to use potassium acetate over sodium acetate because potassium dodecyl sulfate, hence formed is poorly soluble in water. This precipitates and it is efficiently removed.
I wonder if the alkaline lysis extraction would be helped by doing a phenol:chloroform extraction first? Or would that cause a problem?
