What are some techniques where a single promoter/system could be used for activation 20-30 genes without DNA recombination due to same repetitive sequence. Are there any alternative techniques where a single chemical inducer be used for multiple genes?
Depends if you want all the genes being transcribed from a single promoter or multiple copies of the promoter. If you want to do the first option you are probably best off using the T4 self splicing intron to excise CDSs from the transcript. You can also use the 2A translational cleavage signal to allow the separation of polypeptides coded by a single CDS. If you want to have a chemically induced promoter regulate an array of genes just use a Tet On promoter system to transcribe the T7 RNA polymerase (with NLS if for use in eukaryotes) which can then express genes behind a T7 promoter of a defined strength. As T7 promoters are short and you can use variations of the sequence you can reduce the probability of recombination.
