I have got the non specific DNA band with the same primer in 1% agarose gel when DNA is isolated from tissue of chicken rather than blood (But when i use only blood of chicken i have found single specific band?)
What is the reason behind it ?
Please suggest..

Have you checked the od260/280 ratio of the dnas? It is possible that blood dna is higher purity because the cells can be washed and lysed easily but tissue dna may still have inhibitors interfering more with your specific amplification and other impurities or are you looking for a genetic reason like chimaerism at your locusor under the primer ?.
Your question is not completely clear to me.Do you get only the wrong band in tissue or both specific and non specific bands?
I wonder if the sequence of the other band would help identify how it has happened.
No,I am not studying the chimerism.Actually i am geting the both (specific and non specific band ) in case of tissue only.When i isolate the DNA from blood only it gives a specific band.Altough Isolation technique of DNA from tissue and blood are different (phenol chloroform for blood and HotShot for tissues).And till i have not checked the 260/280.
