I am currently culturing Spodoptera frugiperda (Sf9) cells using Grace’s Insect Medium and wanted to clarify whether it is necessary to supplement the medium with L-glutamine for optimal cell growth and protein production.
Since some commercially available formulations of Grace’s Medium do not contain L-glutamine by default, I’m wondering if supplementation is essential when working with Sf9 cells, particularly in suspension culture. If so, what is the recommended concentration, and are there any advantages to using stabilized alternatives like GlutaMAX compared to freshly prepared L-glutamine?
I would appreciate any guidance, shared experiences, or references regarding best practices for glutamine supplementation in insect cell culture using Grace’s Medium.
